Oxygen–dependent regeneration of NADP+ in asymmetric enzymatic oxidations

Alcohol dehydrogenase (ADH) catalyzed oxidations are an appealing concept for obtaining chiral building blocks from symmetric or racemic precursors. However, ADH-catalyzed reactions often exhibit a low turnover in the oxidation direction, which may be due to an unfavorable equilibrium position and/or low enzymatic activity for ADH-catalyzed oxidations compared to reductions.To render ADH-catalyzed oxidations a viable option for biocatalysis, the identification of the molecular reasons for the poor conversion rates would be highly desirable. In this project, we will perform oxidations on different model substrates catalyzed by ADH from Lactobacillus brevis by the use of a novel regeneration system for NAD(P)+. Strategies to improve conversion rates will be developed based on molecular models of substrate/enzyme interactions.
